چکیده
The first monoclonal antibody was appproved for human clinical use in 1985. This antibody, OKT3 (Kung et al., 1979), was derived from a mouse hybridoma and produced anti-mouse reactions that limited its use, and hindered further development of monoclonal antibodies. Humanization technology eliminated these cross-species issues and resulted in the approval of the first recombinant “chimeric” antibody rituximab and “humanized” antibody trastuzumab in 1997 and 1998, respectively. The approval of these molecules opened the floodgates for antibody development, as over 20 antibodies are currently approved by the FDA for therapeutic use and hundreds are in clinical development (nelson et al., yEAR?). The need for humanized recombinant antibodies spurred the develoment of several technologies for their discovery and engineering.

Discovery of Novel Peptidomimetics for Brain-Derived Neurotrophic Factor using Phage Display Technology
Brain-Derived Neurotrophic Factor (BDNF) is a neuroprotectant candidate for neurodegenerative diseases. However, there are several clinical concerns about its therapeutic applications. In the current study, we selected BDNF-mimicking small peptides from phage-displayed peptide library as alternative molecules to the clinical challenges. The peptide library was screened against BDNF receptor (Ne...
متن کاملConstruction of a modular yeast two-hybrid cDNA library from human EST clones for the human genome protein linkage map.
Identification of all human protein-protein interactions will lead to a global human protein linkage map that will provide important information for functional genomics studies. The yeast two-hybrid system is a powerful molecular genetic approach for studying protein-protein interactions. To apply this technology to generate a human protein linkage map, the first step is to construct two-hybrid...
متن کاملA method for global protein expression and antibody screening on high-density filters of an arrayed cDNA library.
We have developed a technique to establish catalogues of protein products of arrayed cDNA clones identified by DNA hybridisation or sequencing. A human fetal brain cDNA library was directionally cloned in a bacterial vector that allows IPTG-inducible expression of His6-tagged fusion proteins. Using robot technology, the library was arrayed in microtitre plates and gridded onto high-density in s...
متن کاملProteome-wide drug screening using mass spectrometric imaging of bead-arrays
A fundamental challenge in the drug discovery process is to develop compounds with high efficacy and minimal side-effects. We describe a new approach to proteome-wide drug screening for detection of on- and off-target binding which combines the advantages of mass spectrometry with microarray technology. The method involves matrix-assisted laser desorption/ionization mass spectrometric imaging (...
متن کاملNegative Regulators of Insulin Signaling Revealed in a Genome-Wide Functional Screen
BACKGROUND Type 2 diabetes develops due to a combination of insulin resistance and beta-cell failure and current therapeutics aim at both of these underlying causes. Several negative regulators of insulin signaling are known and are the subject of drug discovery efforts. We sought to identify novel contributors to insulin resistance and hence potentially novel targets for therapeutic interventi...
